There are many types of ultraviolet-visible spectrophotometers. According to the structure of the instrument, they can be divided into three types: single-beam spectrophotometer, double-beam spectrophotometer and double-wavelength spectrophotometer. Among them, single-beam spectrophotometer and double-beam spectrophotometer belong to single-wavelength spectrophotometer.
classification of uv-visible spectrophotometer:
1, single beam spectrophotometer the light emitted from the light source is split by a monochromator to obtain a beam of monochromatic light. after passing through the absorption cell, it is always a beam of light irradiated on the detector. This type of spectrophotometer has a simple structure and is inexpensive, but its accuracy is poor due to the large influence of stray light, light source fluctuation, etc.
2. The monochromatic light of the composite light emitted by the double-beam spectrophotometer from the light source is divided into two beams of light with equal intensity by the reflector (light cutter) and passes through the reference solution and the sample solution respectively. Since the two beams of light pass through the reference solution and the sample solution at the same time, errors caused by changes in the intensity of the light source can be automatically eliminated. Double beam spectrophotometer can automatically record the absorption spectrum curve, its sensitivity is better, but the structure is more complex, the price is more expensive.
3. The light emitted by the same light source in the dual-wavelength spectrophotometer is divided into two beams, which pass through two monochromators respectively to obtain two beams of monochromatic light with different wavelengths. The two beams of monochromatic light with different wavelengths are alternately irradiated on the same solution at a certain frequency by the light cutter, and then the difference of absorbance at the two wavelengths is obtained through information processing through the photomultiplier tube and the electronic control system. The dual-wavelength spectrophotometry eliminates the background interference and the interference of coexisting components to a certain extent, and improves the sensitivity of the analysis. The dual-wavelength spectrophotometer can be used not only for the analysis of multi-component mixed samples, but also for the analysis of turbid samples (such as biological tissue fluid) that cannot be determined by general spectrophotometers.
